Orient's Gary Alexander netted a last-gasp goal for the second successive week to deny Rochdale a point.
Alexander, whose late strike earned the Londoners a draw against Northampton, produced the winner when he fired home substitute Boniek Forbe's cross.
Matthew Lockwood gave the O's a first-half lead from the penalty spot after Wayne Purser had been grounded.
Dale then levelled via a Lee McEvilly penalty, before Alexander sealed it.
